ABCA1-dependent cholesterol efflux of apoA-I is unclear. Methods and Results - We have proposed that lysine K ; residues in the YxxK motif can direct Tyr chlorination in proteins. To test this hypothesis, we engineered a series of mutations in human apoA-I. When the K residue in the YxxK motif next to Y192 was mutated to arginine, chlorination of Y192 by MPO was blocked. Y166 is resistant to chlorination and does not reside in a YxxK motif. When we introduced the YxxK motif into this region of apoA-I, Y166 was chlorinated in high yield. Importantly, we also found that methionine Met ; residues adjacent to this motif inhibit chlorination, indicating that they can act as local, protein-bound antioxidants. Met residues in apoA-I were oxidized quantitatively by MPO, and this process was completely reversed by treatment with PilB, a methionine sulfoxide reductase. Phenylalanine F ; , in contrast to Tyr, is resistant to chlorination by MPO. Remarkably, when the Y192F mutant of apoA-I was exposed to MPO and then incubated with PilB, its ability to promote cholesterol efflux by the ABCA1 pathway was almost completely restored. Conclusions - These observations strongly support the proposal that YxxK motif directs the regioselective chlorination of Tyr residues in apoA-I. They also argue against the hypothesis that the selective chlorination of Tyr192 requires MPO to interact directly with apoA-I. Our results further suggest that a combination of Y192 chlorination and Met oxidation is both necessary and sufficient for depriving apoA-I of its ABCA1-dependent cholesterol transport activity and malarone.

Biology, Eastern University, St. Davids, PA, USA Toxoplasma parasites and relatives like Plasmodia are sensitive to dinitroaniline herbicides. It is hypothesized that dinitroanilines disrupt parasite microtubules. Our lab and others have demonstrated the effect of dinitroanilines on Toxoplasma replication -- parasites grow, replicate their DNA, but cannot split apart causing parasites to take on a blobbed appearance. Studies on herbicide resistance in goosegrass and green algae identified point mutations in -tubulin genes correlated with resistance to dinitroanilines. Through chemical mutagenesis, we have generated Toxoplasma parasite clones resistant to 1 M benfluralin or trifluralin herbicides. Parasites resistant to benfluralin exhibited a point mutation in nucleotide position 1801 of -tubulin guanine to adenine substitution ; which, when translated, revealed a substitution of valine with methionine at amino acid 252. Likewise, parasites resistant to trifluralin exhibited a point mutation in nucleotide position 1850 of -tubulin resulting in a thymine to cytosine substitution. Translation of this mutated sequence revealed a substitution of methionine with threonine at amino acid 268. These mutations are similar to known mutations that confer resistance to oryzalin another dinitroaniline ; in Toxoplasma or goosegrass. To confirm that these point mutations are responsible for the drug resistance phenotype, the mutated -tubulin gene associated with trifluralin resistance was cloned into a Toxoplasma expression vector and electroporated into wild-type parasites. Transformed parasites were subjected to drug selection using trifluralin ; , and indeed converted from wild-type to the resistant phenotype. Further studies are needed to analyze the mechanism of the insertion of the transgene. Parallel experiments will be performed using the benfluralin-associated mutation. This protist constitutes and ideal model to study the origin and evolution of anaerobic metabolism. Entamoeba histolytica alcohol dehydrogenase 2 EhADH2 ; is a fusion-protein with N-terminal ALDH and C-terminal ADH domains and is essential in glycolytic fermentation of E. histolytica. We have identified a member of the ADHE family from Entamoeba invadens, a reptilian and amphibian pathogen. Expression of EhADH2 and EiADHE restored the ability of a mutant E. coli strain to grow anaerobically. EiADHE showed ADH and ALDH activities with higher resistance to oxygen inactivation. Molecular and biochemical studies demonstrate these bi-functional enzymes require iron for both ADH and ALDH activities. Zinc and chelators dramatically reduced enzymatic activities and anaerobic growth of the recombinant E. coli strains transformed with pEhADH2 and pEiADHE. Addition of iron to E. colipEhADH2 and E. histolytica trophozoites significantly enhanced bacteria and amebic growth while zinc or chelators inhibited or halted it. We are currently testing metal effects on E. invadens trophozoites. These results confirm homology to bacterial alcohol dehydrogenases such as AdhII from Zimomonas mobilis, a "unifunctional" alcohol dehydrogenase. Comparisons between ADHE enzymes human, reptilian, amphibian pathogens or commensal protists will provide insights to their evolutionary history and host immune responses.

This conclusion has been contested 5, 7, 8 ; , and the suggestion has been made that the effect is far more general in scope than previously realized, since growth inhibition is observed even under conditions in which arginine is present in large amounts. Rather, it is claimed that a large pool of one basic amino acid, in this case lysine, simply displaces other basic amino acids, e.g., arginine required for postexponential growth, from the pool. Irrespective of which, if either, of these hypotheses accounts for the absence of an arginine pool, I do not believe that basic amino acid starvation adequately explains postexponential growth inhibition by lysine or any other nonutilizable nitrogen source tested. Although it does seem that protein synthesis is required 1 ; , the size and composition of the basic amino acid pool does not correlate with the presence or absence of postexponential growth on a variety of inhibitors and carbon sources used in the present study. Although D-lysine and 3-alanine were as effective inhibitors as L-lysine in the presence of glucose, the cells did contain arginine in the amino acid pools Table 1 ; . To better understand the relationship between these two Hsp70s, we set out to analyze interactions with the ribosomeassociated nascent chains. Ssb has been crosslinked to ribosomeassociated nascent chains produced in a yeast translation extract 18 ; . However, the chains tested had a limited size distribution. To try to delineate more specifically the sites of interaction of Ssb, mRNAs were produced that encoded pCoxIV polypeptides of 152, 96, 77, or 34 amino acids Fig. 2A ; . These translated chains remained tethered to the ribosome because of the absence of a stop codon. In vitro translation was performed in the presence of [35S]methionine and lysyl-tRNAs charged with lysines bearing a photoactivatable crosslinker, TBDA. Labeled nascent chains bearing this photoactivatable probe can form covalent attachments with interacting proteins, which can be identified by immunoprecipitation. The chains of 152, 96, 77, and 54 amino acids could be crosslinked to Ssb, but we were unable to detect crosslinking to the shortest chain that was 34 amino acids Fig. 2B ; . These results are consistent with Ssb interacting with chains extending beyond the ribosome tunnel, but not those sequestered within it 30, 31 ; . The signal was significantly weaker in the 54-amino acid chain compared with the longer chains. Assuming the ribosome tunnel sequesters 40 amino acids, the only lysine expected to be exposed in this nascent chain would be the lysine at position 12, which would be very close to the exit site. To better understand the interaction of Ssb with the nascent chain, we further characterized the 96-amino acid chain. This polypeptide contains 8 lysine residues, 4 of which would be expected to extend beyond the ribosome tunnel. To assess the contribution of these 4 lysines, at positions 12, 28, 32, and 52, to the crosslinking signal, we made constructs that changed the codons encoding these lysines to those encoding arginines. As expected, if Ssb was being crosslinked to the nascent chain extending beyond the ribosome, no signal was detected if the lysines at positions 12, 28, 32, and 52 were removed. Alterations at positions 12, 28 32, or 52 gave intermediate amounts of and mechlorethamine.

Objective: Recent studies have pointed to the role of the IGF system in adrenocortical tumorigenesis. The IGF-II gene is overexpressed in malignant adrenocortical tumors and its proliferative effects are mediated by the type-1 IGF receptor IGF1R ; . The mannose 6-phosphate IGF2 receptor M6P IGF2R ; plays a key role in regulating cell growth, by ensuring the clearance and inactivation of IGF-II and facilitating activation of the growth inhibitor, transforming growth factor b TGFb1 ; . The M6P IGF2R has been implicated as a tumor suppressor gene in various human tumors. Methods: The purpose of this study was to determine if the M6P IGF2R is involved in adrenal tumorigenesis. Two polymorphisms in the 3 H untranslated region of M6P IGF2R were used to screen a large series of 76 sporadic adrenocortical tumors for loss of heterozygosity LOH ; by PCR amplification of DNA. Tumors were classified into three groups based on pathological features: benign tumors n 25Y suspect tumors n 22 and malignant tumors n 29X Results: LOH at the M6P IGF2R locus was detected in 15 of 26% ; informative tumors and was more frequent in malignant tumors 58% ; than in benign and suspect tumors 9 and 13% respectively ; . Conclusion: These findings provide evidence that LOH at the M6P IGF2R locus is a frequent event in adrenocortical tumors and support the hypothesis that it may function as a tumor suppressor gene in adrenocortical tumorigenesis.
